Effective Purification of Peptides using Normal Phase Flash

20 November 2017

Although linear synthesis of protected peptides is generally straightforward, purification of these compounds using traditional reversed phase methods is quite challenging. A poster from Biotage labs describes the use of normal phase chromatography for purification of fully protected peptides.

As the popularity of post-synthetic peptide chemistry continues to go, additional purification strategies will be required. Standard reversed phase techniques may be ineffecient due to the extreme hydrophobicity imbued by the presence of sidechain protecting groups. Herein, we demonstrate that normal phase chromatography, specifically the use of a methanol:DCM solvent system and a step gradient, can indeed facilitate rapid purification of fully protected peptides.
