Recreational marijuana use is currently legal in eleven U.S. states. Fourteen additional states have decriminalized marijuana, but have not legalized it. It is increasingly important for laboratories to develop accurate testing methods for THC, and its related compounds, in biological matrices of those that may be driving or committing crimes under the influence of marijuana. Biotage provides simple and effective sample prep protocols for cleanup of blood, urine, and oral fluid samples that produce reliable results, and are automation compatible.

Hydroxychloroquine (HCQ) and chloroquine (CQ) are common antimalarial drugs with long half-lives. They are currently being tested as potential treatments for COVID-19. Due to their high publicity in the media, many forensic toxicologists are working on developing methods to extract and detect these compounds in different matrices to understand the adverse effects, potential toxicity, and drug interactions associated with these drugs. These compounds are not typically included in general unknown screens. A simple sample preparation procedure that delivers clean extracts and analyte recovery greater than 90% for CQ and greater than 82% for HCQ with minimum matrix effects and high process efficiency was developed.

LC-MS/MS analysis of volatile analytes is problematic when 96-well plates are used. Solid phase extraction of urine with high drug concentrations can cause contamination of the elution solvent during evaporation as the compounds volatilize then redistribute in adjacent wells. This phenomenon, known as “evaporative crosstalk”, can generate false positive results.

Evaporative crosstalk is well-to-well cross contamination
during extract evaporation after SLE or SPE extraction in a 96
well plate. It is often observed with volatile analytes like
methamphetamine and amphetamine, but it can also occur
with other drugs and metabolites when urine samples have
very high analyte concentrations. ASMS, 2020.

This poster compares the performance of manual processing to a novel automated sample preparation system prior to GC/MS or LC-MS/MS analysis in forensic toxicology applications. Emphasis is placed on the potential for 96-well cross contamination and strategies for its elimination.

This poster demonstrates a rapid and reliable sample preparation method using Supported Liquid Extraction (ISOLUTE SLE+) to extract a suite of 30 hormone analytes from a hydrolyzed urine matrix. Single injection analysis by LC-MS/MS shows that matrix effects are eliminated by the ISOLUTE SLE+ protocol and that analyte recovery and sensitivity have excellent clinical utility.

In all areas of analytical laboratory testing it is vital to ensure proper quality measures are in place to reduce or eliminate cross contamination between samples, which could result in false positive and/or false negative results. In many cases sample carryover in the LC/MS system is checked early on in the method development process. However, one area that can often be overlooked the sample preparation stage. This involves all aspects including pipetting, sample transfer, extraction protocol, evaporation and mixing steps. This poster examines various stages of the sample preparation process to determine the potential for cross contamination and present approaches to minimize and or eliminate the effect. This is demonstrated via a series of dye experiments combined with analyte testing using LC-MS/MS.
