Greening solid phase peptide synthesis: reducing washes while maintaining crude purity

Solid phase peptide synthesis (SPPS) is used in the production of more than 80% of commercially manufactured peptides, and to an even greater extent at smaller scales. With the passing of the REACH Act, use of DMF in chemical synthesis has been restricted heavily, stimulating evaluation of alternative solvent systems and chemistries that enable production of peptides with similar levels of efficiency. Herein, we present data highlighting the impact of washing during SPPS using an automated peptide synthesizer that reduces overall DMF consumption and with the goal to deliver highly pure crude peptides.
