Gel Filtration PhyTip® column

Gel Filtration PhyTip® columns, packed with size-exclusion chromatography resin, provide an automated solution for desalting protein samples and performing buffer exchange. They can be set up as a secondary purification step following affinity PhyTip® columns, for example, to remove imidazole after IMAC purification. 
 

Like other PhyTip® columns, the resin is packed between two inert screens that maintain the resin bed at the base of the column. Unlike other PhyTip® columns, Gel Filtration columns are top-loaded, allowing gravity flow to pull the sample through the gel. Available with 5K or 30K size-exclusion resins, these columns retain small molecules while proteins over 5 kDa or 30 kDa pass through. 
 

Gel Filtration PhyTip® columns support high-throughput processing of antibody and protein samples with automated desalting and buffer exchange. They remove >95% of salts while maintaining >80% protein yield and preserve protein functionality. Results are highly reproducible, with <10% CVs across sample volumes and concentrations. 
 

Up to 96 samples can be processed in approximately 30 minutes, much faster than traditional overnight dialysis. Gel Filtration PhyTip® columns are compatible with any liquid handling system with a gripper arm, which can move trays for desalting, buffer exchange, or fraction capture during 96-sample screening runs.
