Sample preparation is necessary to ensure that the analytical determination system evaluates only those substances that are strictly compatible with the analytical techniques used, thus avoiding false negatives (analytes present in the sample are not detected), or false positives (interferences in the sample are taken to be signals from the expected analyte).

Similarly, components of interest may only be present in trace amounts, meaning an extract needs to be concentrated for the components to be detected. These are reasons why a sample preparation process is required for most analytical determination systems handling biological fluids and organic samples.
