Fast, automated purification of His-tagged recombinant proteins.
IMAC PhyTip® columns enable automated purification of histidine-tagged proteins, delivering high recovery and purity while being gentle on the protein. Recombinant proteins are often expressed with affinity tags to simplify purification, with the histidine (His) tag being the most common. This tag, typically composed of six or more histidine residues, is purified using immobilized metal affinity chromatography (IMAC) resins, with mild elution achieved using buffers containing 100–250 mM imidazole.
Packed with high-capacity Nickel-IMAC resin, IMAC PhyTip® columns purify His-tagged recombinant proteins with 60–80% recovery and routinely produce samples with >90% purity. PhyTip® columns use dual-flow chromatography technology to ensure efficient binding of the entire sample to a small resin bed, enabling elution in small volumes and generating highly concentrated protein samples.
IMAC PhyTip® columns are available in resin bed volumes from 5 μL to 320 μL, suitable for protein samples from microgram to milligram scale and sample volumes between 100 μL and 50 mL. They are compatible with all major liquid handling platforms, allowing integration with existing lab instrumentation, and come with pre-programmed automation protocols that can be easily customized for your application.
