This report details a “load, wash, elute” weak cation exchange solid phase extraction procedure amenable to both plasma and urine samples. The extracts are subsequently injected into an LC-MS/MS system. The preliminary sample preparation method was developed at the Biotage US Applications lab (Charlotte, NC).The method was then transferred to Ionics (Bolton, ON, Canada) to facilitate the nmole/L measurements of the selected biomarkers
by laminar flow tandem mass spectrometry. The SPE-LC-ESIMS/ MS method parameters were first optimized using pooled mixed gender plasma. A set of patient samples (n=32) was later supplied by the Mayo Clinic (Rochester, MN) that had previously been analyzed by a validated referee method. The population represented measured values across a range of clinical relevance.

In this poster, an IONICS 3Q 220 triple quadrupole mass spectrometer using a load, wash, elute method protocol with recently advanced solid phase extraction (SPE) sorbent technology (EVOLUTE EXPRESS WCX) was used to perform analysis of the two compounds. As a result low levels of metanephrine and normetanephrine are detectable in plasma with short sample preparation and LC run times. This LC-MS/MS method provides a fast, sensitive, accurate, and reproducible solution for the analysis of ME and NME in plasma.

We detail here the application of new column technology to purify histamine and serotonin (Table 1) in one extraction method with the elimination of the sorbent conditioning and equilibration steps familiar to traditional solid phase extraction (SPE) method protocols prior to measurements by ultra performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-MS/MS). A secondary goal of combining two LC methods into one method was also achieved.

This technical note provides guidelines for the extraction of quaternary amines and strongly basic analytes from aqueous samples using EVOLUTE WCX (Method A). A method optimized for extraction of a wide range of basic compounds, allowing elution under mildly acidic conditions (Method B) is also detailed. This approach is recommended for the extraction of basic compounds when basic elution conditions are unsuitable, or acidic elution conditions are preferred due to LC-MS/MS compatibility. The procedures are described on page 1–2, with processing and optimisation guidelines on page 3. An example application showing the extraction of quaternary amines from urine and plasma illustrates the use of EVOLUTE WCX (Chlormequat, Neostigmine, Bretylium, Difenzoquat) for clean-up of biological fluid samples.
