Ion exchange chromatography can be used either for a separate purification step or for a second polishing purification step after affinity chromatography purification as an orthogonal chemistry to obtain a highly pure final product. The result is complete protein capture and high final protein concentrations. Due to the physiological buffers used during the protocol, ion exchange chromatography is a generally gentle purification method.

Biotage offers ion exchange PhyTip columns with different types of ion exchange resin for easy customization of your purification protocol. Cation exchange separations are typically performed at pH between 4 - 7 and anion exchange separation at pH 5 -8. Within anion exchange there are two types: strong base anion exchange and weak base anion exchange. The same applies to cation exchange: there is strong acid cation exchange and weak acid cation exchange.
Our PhyTip® columns are available for all major liquid handling systems and in resin bed volumes from 5 μL to 320 μL for processing of a wide range of samples.
