Unbiased screening technologies have opened the door to expanded repertoires of peptides that can quickly and easily be screened in early lead identification programs, making this a much more attractive modality in a drug discovery pipeline. Once hit compounds are identified in the preliminary screen, secondary peptide synthesis for validation and preliminary structure-activity relationship (SAR) characterization is readily achieved with high throughput automated peptide synthesis platforms.

    However, traditional downstream processing
approaches introduce a variety of bottlenecks that
slow delivery of the compound library for evaluation
in downstream assays. An alternative strategy, Solid
Phase Extraction (SPE), can be used to increase the
purity of crude peptide libraries to an acceptable
level for preliminary activity assays with predictable
recovery behavior in an expeditious, parallel and
    automated manner. When combined, automated
high throughput synthesis and SPE-based cleanup
enable the most efficient peptide library delivery for
    downstream evaluation.
